抗體-科研偶聯(lián)物(ADCs)是一類重要的靶向生物科研。ADCs由單克隆抗體和細(xì)胞性科研(小分子化)組成,通過(guò)半、賴氨酸或其他工程化殘基,通過(guò)多種連接子(linker)連接在一起。因此,可以實(shí)現(xiàn)ADC作為化療科研的性遞送至細(xì)胞,從而靶位點(diǎn)的藥理活性,同時(shí)使非靶免受化療損傷,由于ADCs的結(jié)構(gòu)復(fù)雜,其穩(wěn)定性要求過(guò)了分子各部分的簡(jiǎn)單總和,因?yàn)閱慰寺】贵w、小分子化和連接子的穩(wěn)定性對(duì)ADCs的性、和潛在的科研抗體(ADA)反應(yīng)都有影響,此外,由于其小分子化的獨(dú)*力,ADCs在性、性和穩(wěn)定性方面滿足一些苛刻的標(biāo)準(zhǔn),以實(shí)現(xiàn)將細(xì)胞性科研且選擇性地遞送至靶點(diǎn)。? 小分子化通過(guò)連接子與抗體偶聯(lián),連接子是ADC的重要組成部分。連接子可以是不同類型的,例如腙(hydrazone)連接子、二硫化物(disulfide)連接子和斷裂或非斷裂的肽連接子。這些連接子釋放活性小分子化的方式在體內(nèi)有的不同。腙連接子對(duì)pH敏感,在較低pH下,通過(guò)在包涵體和/或溶酶體等隔間中斷裂釋放活性小分子化。二硫鍵利用細(xì)胞內(nèi)的還原環(huán)境釋放小分子化。已知斷裂的肽連接子被溶酶體蛋白酶酶切,而非斷裂的連接子通過(guò)溶酶體隔間中抗體的降解釋放活性小分子化。近發(fā)現(xiàn)基于肽的連接子(斷裂和非斷裂)更常用,因?yàn)樗鼈冿@示出更高的血漿穩(wěn)定性和更高的活性??傮w而言,由于代ADC中使用了化學(xué)不穩(wěn)定的肼和某些二硫鍵,通過(guò)創(chuàng)建酶促斷裂和非斷裂的連接子,連接子穩(wěn)定性得到了。二硫鍵的化學(xué)性質(zhì)也隨著保護(hù)基團(tuán)的引入而得到,例如,鏈間二硫鍵的減少,隨后加入含有炔基的二硫代苯基馬來(lái)酰亞胺鍵,使得能夠與含有疊氮化物基團(tuán)的小分子化偶聯(lián)。? (如二肽型可切割連接蛋白),則溶酶體酶(如蛋白酶B)能夠性地切割連接蛋白,釋放小分子化用于LC-MS檢測(cè),這是在抗性mAb親和捕獲法從基質(zhì)中捕獲ADCs后實(shí)現(xiàn)的。如果連接子不可斷裂,而該酶不能從連接子上斷裂小分子化,那么在從基質(zhì)中捕獲抗性單克隆抗體后,將應(yīng)用抗體的蛋白酶(例如胰蛋白酶)消化,并且肽偶聯(lián)的小分子化是由LC-MS/MS檢測(cè)的替代分析物(圖1)。基于抗性單克隆抗體捕獲的LC-MS/MS分析能夠檢測(cè)與單克隆抗體偶聯(lián)的小分子化。然而,如果使用抗小分子化單克隆抗體作為捕獲試劑,可檢測(cè)樣品中潛在的類型小分子化分子,包括未偶聯(lián)的小分子化、與單克隆抗體偶聯(lián)的小分子化、與連接子偶聯(lián)的小分子化以及與小分子化偶聯(lián)的單克隆抗體的分解代謝物。
